Hair follicles play an important role in hair development. This study aimed to develop a microgel-spotting device fabricate multilayered gel bead culture model and mimic the early development of skin appendages regenerate vitro. The consists alginate layer containing cytokines as core layer, collagen mouse embryonic stem cells middle fetus-derived epidermal outer layer. A concentration gradient is formed, which promotes interactions between cells. Histological immunnohistological analyses confirmed reconstruction follicle structures. As result, cell number size could be precisely controlled by developed device. In bead, cultured with cytokine formed aggregates keratinized tissue center similar “native” structure. Sweat gland-like luminal erector pilorum-like structures were also observed around concentric conclusion, demonstrated potential for vitro regeneration. findings this provide insight into appendages.
